The Plant Allergens group (alislab151) is studying molecular basis of allergy for over 20 years, characterizing plant allergens, and improving their diagnosis and treatment. Our research follows two complementary strategies combining techniques and methods from Biochemistry, Molecular Biology, and Immunology to identify and characterize the molecular mechanisms of the allergic response, with computational methods to study the physicochemical and structural properties of allergens. Docking analyses, Molecular Mechanic Calculations and Molecular Dynamics simulations are employed to model protein-ligand binding and oligomerization states of allergens.
Why do we work on food allergy in a plant center like the CBGP?
Because many plant allergens are plant defense proteins (Table). Therefore, modified plants, more resistant to climate change, with less nutritional requirements, etc., might increase the quantity of allergens, with the concomitant risk for public health.
Plant lipid transfer proteins (LTPs) are the major allergens in the Mediterranean area
Working with five allergenic LTPs (Pru p 3 (as model), Tri a 14, Art v 3, Par j 2, and Ole e 7), our lab demonstrated that they transport a ligand with the same chemical identity. Further characterization of the ligand showed the role of the polar head and phytosphingosine tail in sensitization, being a critical factor in this process.
To show the immunological role of the LTPs’ ligand, two non-LTP-proteins, bovine serum albumin (BSA) and ovalbumin (OVA), were used in an allergic mouse model in complex with it. Computational analysis predicted BSA to bind five molecules of LTPs’ ligand, and OVA, only bound (A), which was confirmed in-vitro (B). Both complexes (BSA-ligand and OVA-ligand) promoted immune response on THP1-XBlue cells (C).
Moreover, our lab developed allergic mouse models using two different strains (C3H and BALB/c) exposed to both complexes as skin sensitizers (D). Only mice sensitized with Protein-LTPs’_ligand complex suffered a drop in temperature after challenge (E), confirming the relevance of the LTPs’ ligand in the allergic sensitization process. |
Improving the food allergy diagnosis: Point-of-care device
The AllerScreening project (allersecreening.com) has focused on translating an optical diagnostic technology and its read-out-platform (MOX), to the clinical routine, addressing a healthcare need. This novel, multiplexed, innovative, cost-effective system for the diagnosis of food allergies help clinicians to detect food allergies through a simple test using sera by a competitive cost. The technology developed is positioned in a TRL6, that is, the AllerScreening system has been proved in a real scenario like the final ones in case it will be upgraded to TRL9.
Working to prevent unwanted encounters: searching for hidden allergens
The search for definitive treatments for food allergy
In the search for a food allergy treatment, our lab makes use of VLPs by coupling TuMV-derived VLPs to Pru p 3. VLPs-Pru p 3 are produced in Nicotiana benthamiana plants and analyzed by EM and DLS (Figure).
The IT-formula was studied as therapy in a mouse model of peach allergy (A). Sublingual administration of the IT-formula reduced serological markers associated with allergic responses in 30% of the mice, such as anti-Prup3_sIgE (B-D), and reduced leukocyte infiltration in treated mice to similar levels as controls (E-F). Noteworthy, no associated macroscopic, nephritic, or hepatic toxicity was detected, showing the safety of the IT-formula. Our lab keeps developing and optimizing the formula in the search of an effective treatment of food allergy.
