Better in tandem

Over recent years, our group worked on the crosstalk between two, on the first sight, antagonistic plant hormones, the major growth promoting auxin indole-3-acetic acid (IAA) and the stress hormone jasmonic acid (JA). Our work demonstrated that wounding triggers the local formation of JA which, in turn, leads to an induction of auxin biosynthesis around the wound site. However, the molecular mechanism by which JA-signaling connects with auxin biosynthesis remained largely unclear. In the presented work, we addressed the question as to whether the major transcription factors driving JA-responses, MYC2, MYC3, and MYC4, are involved in the transcriptional regulation of two key players in auxin biosynthesis, YUC8 and YUC9. Among other things, our investigations highlighted a characteristic tandem G-box motif, consisting of two canonical G-boxes (5’-CACGTG-3’) and one G-box variant (5’-CACGTC-3’), that is likely responsible for the JA-mediated induction of the two YUCCA genes. In addition, our work highlighted that YUC9 plays a seemingly important role in plant defense responses towards biotic foes. Testing the feeding behavior of spider mites (Tetranychus urticae) on leaves of wild-type and YUC9 mutant plants clearly suggested that the overexpression of YUC9 translates into reduced tissue damage, because the spider mites preferred to leave the YUC9oe leaves. With respect to our previous results, this is likely due to the induced lignification of tissues that overexpress YUC9.
